<Lecture 1: Aiming to Contribute to Medicine through Optogenetics: Research on Rhodopsin Responsive to Violet to Near-Infrared Light>
Optogenetics is a technology to manipulate biological functions by expressing photoreceptor proteins such as rhodopsin in specific cells. By developing photoreceptor molecules with various characteristics, this technology can be applied to the elucidation of all kinds of biological phenomena. In this presentation, we will introduce an example of neural activation by rhodopsin, a channel that absorbs violet light and has high calcium permeability, and the molecular mechanism of rhodopsin, an enzyme that absorbs near-infrared light and shows cyclase activity, recently discovered by our research group.

Recent breakthroughs with in vitro transcribed mRNA technology have allowed it to emerge as a promising solution for therapeutic obstacles. The launch of the two mRNA-based SARS-CoV-2 vaccines demonstrated key advantages of the technology including rapid development, easy scaleup of production, and robust in vivo expression. The stability, function, and immunogenicity of a given mRNA is affected by many intrinsic factors including untranslated regions, coding sequence, polyA tail, production conditions (modified nucleotide incorporation, purification), etc. VectorBuilder Inc. generates mRNA, and its efficacy has been thoroughly validated both in vitro and in vivo. We provide one-stop service from design to production to help researchers develop novel therapeutics using this exciting technology.

<Lecture 3: Analysis of various AAVs using adeno-associated virus (AAV) receptor-immobilized columns (tentative)>
Adeno-associated virus (AAV) vectors are widely used as gene therapeutic agents. The authors immobilized the AAV receptor (AAVR), which contributes to AAV infection, on a gel, developed an affinity chromatography column, and chromatographically analyzed various types of AAV. The elution times of AAV produced by us and VectorBuilder were identical for each serotype. In addition, AAV4, which does not have AAVR-dependent infection, was passed through the chromatogram, suggesting that analysis close to the intracellular environment is possible.
